- GBP2 (Guanylate-binding protein 2, also known as GBP-2 or HuGBP-2) is an interferon-inducible GTPase that belongs to the guanylate-binding protein family.
- It hydrolyzes GTP to GMP in two consecutive cleavage reactions, with GDP as the major product. Related references: PMID:8706832
- GBP2 plays crucial roles in innate immunity against bacterial, viral, and protozoan pathogens by targeting pathogen-containing vacuoles.
- Upon infection, GBP2 is recruited to vacuoles or escaped bacteria, promoting vacuolar lysis and release of pathogens into the cytosol, which enables detection by inflammasomes.
- It facilitates the release of bacterial ligands like LPS and dsDNA, activating non-canonical CASP4/CASP11 and AIM2 inflammasomes, respectively.
