Biological Background: Fatty Acid Synthase Function and Localization

  • Fatty acid synthase (FASN) is a large multifunctional enzyme that catalyzes the de novo biosynthesis of long-chain saturated fatty acids from acetyl-CoA and malonyl-CoA using NADPH.
  • The enzyme contains seven distinct catalytic activities and an acyl carrier protein (ACP) domain with a 4'-phosphopantetheine prosthetic group.
  • FASN is a key enzyme in lipid metabolism and is ubiquitously expressed, with prominent levels in brain, lung, liver, and mammary gland.
  • Subcellularly, FASN localizes to the cytoplasm and has also been detected in melanosomes.
  • FASN activity is required for the replication of SARS-CoV-2, indicating a role in viral infection.
  • Post-translational modifications include acetylation, phosphorylation, S-nitrosylation, and isopeptide bond formation, which may regulate its function.
  • As a member of the short-chain dehydrogenase/reductase superfamily, FASN belongs to the SDR27X1 subfamily (also known as FAS, OA-519).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 2212–2511 of human FASN, which lies within the terminal region of the enzyme; consider verifying specificity in your assay system.
  • For Western blot (WB), a band at approximately 273 kDa is expected; use an appropriate high-molecular-weight gel and transfer conditions.
  • In immunofluorescence (IF/ICC), FASN shows a cytoplasmic distribution; optimize permeabilization and fixation protocols for your cell type.
  • For ELISA applications, confirm antibody pair compatibility and adjust coating and detection conditions as needed.
  • Validate cross-reactivity in mouse and rat samples, as the antibody is predicted to recognize FASN from these species.
  • Consider using this antibody to study fatty acid metabolism, cancer metabolism, or viral replication pathways.
