Biological Background: Gasdermin E Function and Localization
- Gasdermin E (GSDME), also known as DFNA5 or ICERE-1, is a member of the gasdermin family of pore-forming proteins. It is expressed as a precursor that is cleaved to release an active N-terminal fragment, with multiple isoforms produced by alternative splicing.
- Upon cleavage by caspase-3 (CASP3) or granzyme B (GZMB) at Asp270, the N-terminal fragment translocates to the plasma membrane, oligomerizes, and forms pores of 10–15 nm inner diameter, triggering pyroptosis and the release of inflammatory cytokines such as IL-1β and IL-16.
