  • Cyclin-dependent kinase 5 (CDK5) is a proline-directed serine/threonine-protein kinase, also known as cell division protein kinase 5, tau protein kinase II catalytic subunit, or PSSALRE.
  • CDK5 is essential for neuronal cell cycle arrest and differentiation, and plays critical roles in neuronal survival, migration, axonal and neurite growth, synaptogenesis, and synaptic plasticity.
  • The kinase is activated by binding to regulatory subunits CDK5R1 (p35) and CDK5R2 (p39), primarily in postmitotic neurons, and phosphorylates a wide range of substrates including MAPT/TAU, MAP2, p53, HDAC1, and small GTPases.
  • In the nucleus, CDK5 phosphorylates transcription factors such as MEF2, p53, and CLOCK, regulating gene expression, apoptosis, and circadian rhythms.
