Biological Background: c-Myc Function and Localization

  • c-Myc (also known as MYCC, bHLHe39, MRTL) is a proto-oncogene transcription factor belonging to the basic helix-loop-helix (bHLH) family, encoded by the MYC gene.
  • It binds DNA specifically to the E-box sequence 5'-CACGTG-3' and non-specifically, activating transcription of growth-related genes including VEGFA to promote angiogenesis. Related references: PMID:24940000 PMID:25956029
  • c-Myc also regulates pre-mRNA splicing by positively controlling expression of HNRNPA1, HNRNPA2, and PTBP1, which in turn modulate PKM alternative splicing. Related references: PMID:20010808
  • Multiple isoforms are produced through alternative initiation and alternative splicing, contributing to functional diversity.
  • It localizes to the nucleus (nucleoplasm, nucleolus), cytoplasm, and chromosomes, consistent with its roles in transcription and chromatin regulation.
  • Post-translational modifications include acetylation, phosphorylation, ubiquitination (isopeptide bond), and O-glycosylation, which regulate its stability and activity.
  • As a key regulator of cell proliferation and self-renewal, c-Myc is frequently overexpressed or amplified in a wide range of human cancers, making it a prominent target for diagnostic and research applications.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning the N-terminal amino acids 1–100 of human c-Myc (P01106); reactivity may cover full-length and select isoforms.
  • For Western blot, a band around 51 kDa is expected; include appropriate loading controls and consider cell treatment to modulate c-Myc expression.
  • For ChIP and CUT&Tag, optimize antibody concentration and assay conditions (e.g., crosslinking, enzymatic digestion) with known target and non-target loci.
  • Since cross-reactivity is indicated for human, verify species compatibility if studying other organisms.
  • KO validation confirms specificity; nonetheless, confirm detection in relevant positive and negative control samples.
