Biological Background: TRIM25 Function and Localization
- TRIM25, also known as EFP, RNF147, or ZNF147, is an E3 ubiquitin/ISG15 ligase belonging to the tripartite motif (TRIM) protein family. Related references: PMID:16352599
- It serves as a critical mediator of innate antiviral immunity by catalyzing Lys-63-linked polyubiquitination of the viral RNA sensors RIG-I and IFIH1, leading to interferon production. Related references: PMID:17392790, PMID:29357390, PMID:30193849, PMID:23950712
- TRIM25 additionally regulates diverse substrates, including 14-3-3 sigma (SFN) via ISGylation, ZFHX3 via proteasomal degradation, and the antiviral factor ZAP/ZC3HAV1 via ubiquitination to enhance mRNA binding. Related references: PMID:16352599, PMID:17069755, PMID:22452784, PMID:28060952, PMID:28202764
- In the nucleus, TRIM25 promotes restart of stalled DNA replication forks by ubiquitinating the BLM helicase, mediated by the KHDC3L-OOEP scaffold.
- TRIM25 suppresses autophagy by ubiquitinating and degrading the cargo receptor p62/SQSTM1. Related references: PMID:40542578
- The protein localizes predominantly to the cytoplasm, but also accumulates in stress granules and the nucleus.
- TRIM25 is ubiquitously expressed, with elevated protein levels detected in breast tumors.
- It undergoes acetylation and phosphorylation and contains characteristic RING, B-box, and coiled-coil domains.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 100–200 of human TRIM25. This region is expected to be accessible and may provide good epitope recognition; however, validate the antibody in your specific sample type.
- This antibody is validated for Western blot (WB) and ELISA applications. For WB, consider using positive control lysates from human cell lines or tissues expressing TRIM25. Due to KO validation, it likely exhibits high specificity; confirm knockdown efficiency in your system.
- Given TRIM25's involvement in innate immunity, treatment of cells with viral mimics (e.g., poly(I:C)) or interferons may modulate expression levels, which can be assessed by this antibody.
- For ELISA, consider using recombinant TRIM25 protein or cell lysates as coating antigen. Optimize antibody dilution based on specific assay conditions.
