Biological Background: TRIM21 Function and Localization

  • TRIM21 (also known as Ro52, SSA1, RNF81) is a 54 kDa E3 ubiquitin-protein ligase and a member of the tripartite motif (TRIM) family, characterized by RING finger, B-box, and coiled-coil domains.
  • It functions as an E3 ubiquitin ligase dependent on E2 enzymes UBE2D1, UBE2D2, UBE2E1, and UBE2E2, forming a complex with UBE2D2 that mediates self-ubiquitination and ubiquitination of substrates such as USP4 and IKBKB.   • TRIM21 is a component of cullin-RING-based SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complexes, such as SCF(SKP2)-like complexes, which ubiquitinate CDKN1B phosphorylated at Thr-187, promoting its proteasomal degradation and regulating cell cycle progression.
