Biological Background: TLK2 Function and Localization
- TLK2 (Tousled-like kinase 2) is a serine/threonine-protein kinase involved in chromatin assembly, DNA replication, transcription, repair, and chromosome segregation (PubMed:10523312, PubMed:11470414, PubMed:12660173). Related references: PMID:10523312, PMID:11470414, PMID:12660173
- It phosphorylates the chromatin assembly factors ASF1A and ASF1B; phosphorylation of ASF1A prevents its proteasome-mediated degradation, thereby enhancing chromatin assembly (PubMed:11470414, PubMed:20016786, PubMed:29955062). Related references: PMID:11470414, PMID:20016786, PMID:29955062
- TLK2 acts as a negative regulator of amino acid starvation-induced autophagy (PubMed:22354037). Related references: PMID:22354037
- The protein localizes to the nucleus (nucleoplasm), as well as the cytoplasm (perinuclear region and cytoskeleton).
- Tissue expression is broad, with highest levels in testis, and moderate expression in placenta, fetal liver, kidney, pancreas, heart, skeletal muscle, spleen, thymus, colon, ovary, small intestine, prostate, and peripheral blood leukocytes; almost undetectable in adult liver and lung.
- Keywords associated with TLK2 include cell cycle, DNA damage, chromatin regulator, and phosphoprotein, highlighting its role in cell division and genome stability.
- The protein contains an ATP-binding domain, a coiled-coil region, and is subject to alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 1-220, encompassing the N-terminal kinase domain of TLK2. Consider epitope mapping if precise binding details are required.
- For western blot, the expected molecular weight is approximately 88 kDa. Verify band specificity using knockout or knockdown controls in the appropriate cell lines.
- The antibody is validated for WB and ELISA. For other applications like IHC or IF, optimization is recommended.
- Cross-reactivity is reported with human and mouse TLK2. For other species, sequence alignment may predict reactivity, but experimental validation is necessary.
