Biological Background: Smad3 Function and Localization
- SMAD family member 3 (SMAD3), also known as Mothers against decapentaplegic homolog 3 (MADH3) or hMAD-3, is encoded by the SMAD3 gene and functions as a receptor-regulated SMAD (R-SMAD) in the TGF-beta signaling pathway.
- Acts as an intracellular signal transducer and transcriptional modulator: upon activation by TGF-beta or activin type 1 receptor kinases, it forms a complex with SMAD4 and translocates to the nucleus to regulate target gene transcription.
- Binds to the TRE element in promoters of TGF-beta-responsive genes and can form a SMAD3/SMAD4/JUN/FOS complex at AP-1/SMAD sites to fine-tune transcription.
- Shuttles between the cytoplasm and nucleus; predominantly cytoplasmic in the absence of ligand and accumulates in the nucleus upon pathway activation (PMID: not available).
- Regulates chondrogenesis and osteogenesis, inhibits early bone fracture healing, and modulates wound healing by affecting keratinocyte growth/migration and monocyte chemotaxis in a hormone-sensitive manner.
- Subject to multiple post-translational modifications, including phosphorylation, acetylation, ADP-ribosylation, and ubiquitination, which control its activity, stability, and interactions; it also contains zinc-binding domains essential for DNA binding.
- Multiple isoforms are produced by alternative splicing, contributing to functional diversity.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 100-200 of human Smad3 (NP_005893.1). This region is conserved, explaining the antibody’s cross-reactivity with mouse and rat Smad3.
- For Western blot (WB), a band at approximately 48 kDa is expected. Include proper loading controls and molecular weight markers to confirm band size.
- For immunohistochemistry (IHC-P), antigen retrieval conditions (e.g., heat-induced epitope retrieval) may require optimization based on tissue type and fixation method.
- The antibody has been validated for chromatin immunoprecipitation (ChIP); using positive and negative control loci for Smad3 binding is recommended to ensure specificity.
- Also suitable for immunoprecipitation (IP) and ELISA. End-users should titrate the antibody to determine the optimal dilution for their specific experimental systems.
