Biological Background: SIRT3 Function and Localization
- SIRT3 (SIR2L3, T3) is a mitochondrial NAD-dependent protein deacetylase with additional protein-lysine delactylase and debenzoylase activities. Related references: PMID:12186850, PMID:12374852, PMID:16788062
- It deacetylates key mitochondrial metabolic enzymes including ACSS1, IDH, GDH, SOD2, PDHA1, LCAD, SDHA, MRPL12, and ATP5PO, thereby regulating their activity. Related references: PMID:16788062, PMID:18680753, PMID:19535340
- SIRT3 plays a central role in cellular energy metabolism and maintaining tissue-specific ATP levels. Related references: PMID:24252090, PMID:18794531
- Under metabolic stress, SIRT3 deacetylates FOXO3, promoting its recruitment to mtDNA with POLRMT to increase mtDNA transcription. Related references: PMID:23283301
- SIRT3 regulates ceramide metabolism by deacetylating ceramide synthases (CERS1/2/6) and influences hepatic lipogenesis via the SLC25A47-AMPK axis.
- It catalyzes delactylation of histone H4 at Lys-16 and CCNE2, and debenzoylation of PPIF and ACLY. Related references: PMID:39524354, PMID:36896611, PMID:37720100
- SIRT3 is localized in the mitochondrial matrix and is widely expressed across human tissues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 300-399) of human SIRT3, which may detect both full-length and processed forms. Validate in your sample system to confirm specificity.
- For Western blot, a band at approximately 44 kDa is expected. Optimize antibody dilution and blocking conditions for your samples.
- This antibody is validated for ELISA; it may be used as a capture or detection reagent after appropriate pairing and validation.
- Cross-reactivity has been tested only in human; if working with other species, verify reactivity experimentally.
