  • Rho GDP-dissociation inhibitor 1 (RhoGDI, also known as Rho-GDI alpha) is encoded by the ARHGDIA (GDIA1) gene and is a key regulator of Rho GTPase signaling.
  • It inhibits the dissociation of GDP from Rho proteins such as CDC42, RAC1, and RHOA, thereby maintaining them in an inactive cytosolic pool and protecting them from degradation.
  • RhoGDI actively participates in the recycling and distribution of activated Rho GTPases; its exceptionally high affinity for prenylated forms allows it to extract both inactive and activated molecules from membranes.
  • Through modulation of Rho proteins, RhoGDI may regulate cell motility; in glioma cells, it inhibits migration and invasion by mediating SEMA5A/PLXNB3 signals that inactivate RAC1.
  • Subcellular localization is predominantly cytoplasmic.
