Biological Background: RELA Function and Localization
- RELA, also known as transcription factor p65 or nuclear factor NF-kappa-B p65 subunit, is a member of the NF-kB/Rel protein family.
- NF-kB is a pleiotropic transcription factor that regulates biological processes such as inflammation, immunity, differentiation, cell growth, tumorigenesis, and apoptosis.
- RELA forms homo- or heterodimers with other Rel-like domain-containing proteins; the RELA-NFKB1 heterodimer is the most abundant and functions as a transcriptional activator.
- NF-kB dimers are retained in the cytoplasm in an inactive state by binding to IkB inhibitors; upon stimulation, IkB is phosphorylated and degraded, allowing NF-kB to translocate to the nucleus.
- Subcellular localization is dynamic: predominantly cytoplasmic in resting cells and nuclear upon activation; it shuttles between the nucleus and cytoplasm.
- Post-translational modifications, including acetylation, phosphorylation, and methylation, modulate its transcriptional activity and interactions.

- The immunogen is a recombinant protein covering mouse RELA; the antibody is polyclonal and validated by knockout (KO).
- For Western blot, expect a band at ~60 kDa. Use appropriate positive controls (e.g., mouse tissue lysates known to express RELA) and validate specificity in your sample system.
- For ELISA, consider checkerboard titration to optimize coating and detection antibody concentrations; cross-reactivity with human allows potential use in human samples after validation.
- Due to the dynamic regulation of RELA, consider using both cytoplasmic and nuclear fractionation to assess activation status.
