Biological Background: RELA Function and Localization

  • RELA, also known as transcription factor p65 or nuclear factor NF-kappa-B p65 subunit, is a member of the NF-kB/Rel protein family.
  • NF-kB is a pleiotropic transcription factor that regulates biological processes such as inflammation, immunity, differentiation, cell growth, tumorigenesis, and apoptosis.
  • RELA forms homo- or heterodimers with other Rel-like domain-containing proteins; the RELA-NFKB1 heterodimer is the most abundant and functions as a transcriptional activator.
  • NF-kB dimers are retained in the cytoplasm in an inactive state by binding to IkB inhibitors; upon stimulation, IkB is phosphorylated and degraded, allowing NF-kB to translocate to the nucleus.
  • Subcellular localization is dynamic: predominantly cytoplasmic in resting cells and nuclear upon activation; it shuttles between the nucleus and cytoplasm.
  • Post-translational modifications, including acetylation, phosphorylation, and methylation, modulate its transcriptional activity and interactions.
