Biological Background: Rad23A Function and Localization
- Rad23A, also known as HR23A or hHR23A, is a multifunctional protein that is also referred to as lysine-specific demethylase RAD23A.
- Functions in histone H4K20 demethylation, removing mono-, di-, and trimethyl marks to produce unmethylated H4K20, thereby regulating transcriptional activation of coding genes and repetitive elements. Related references: PMID:32209475
- Participates in nucleotide excision repair (NER) by forming a complex with XPC; the XPC-RAD23A complex is critical for global genome NER and can stabilize XPC. Related references: PMID:9372924
- Involved in ubiquitin-dependent protein degradation by binding to 'Lys-48'-linked polyubiquitin chains and delivering ubiquitinated substrates to the 26S proteasome. Related references: PMID:12643283 PMID:14621999 PMID:15321727
- Plays a role in HIV-1 replication by facilitating the association of HIV-1 Vpr with the host proteasome in non-proliferating cells and primary macrophages.
- Localizes predominantly to the nucleus, consistent with its functions in DNA repair, chromatin modification, and transcriptional regulation.
- Contains ubiquitin-like (UBL) domains and belongs to the UBL family; keywords include Ubl conjugation, repeat, and phosphoprotein.
Experimental Guidance and Technical Tips
- This rabbit polyclonal antibody was generated against a recombinant protein encompassing amino acids 1-363 of human Rad23A, which includes N-terminal ubiquitin-like domains and functional regions. Consider validating specificity using knockout (KO) cell lines or tissues, in line with the antibody's KO-validated status.
- For Western blot analysis, a band at approximately 40 kDa is expected. Due to potential post-translational modifications or isoform expression, confirm the observed molecular weight in your specific sample system.
- The antibody is suitable for ELISA applications; optimize coating conditions and antibody dilutions for your assay format.
- Cross-reactivity has been demonstrated with human and mouse samples; if using other species, perform appropriate verification.
