Biological Background: PML Function and Localization
- Encoded by the PML gene, Protein PML is also known as E3 SUMO-protein ligase PML, Promyelocytic leukemia protein, RING finger protein 71, and Tripartite motif-containing protein 19 (TRIM19). It belongs to the TRIM family and contains a RING finger domain, B-box, coiled-coil region, and a C-terminal domain subject to alternative splicing, generating multiple isoforms with distinct functions.
- Acts as the essential scaffold protein of PML nuclear bodies (PML-NBs), dynamic subnuclear structures that regulate tumor suppression, transcriptional control, apoptosis, senescence, DNA damage responses, and antiviral defense by recruiting and modulating numerous partner proteins via SUMO-mediated interactions.
- Inhibits EIF4E-mediated mRNA nuclear export by reducing EIF4E binding affinity for the 5' m7G cap of target mRNAs, thereby controlling gene expression at the post-transcriptional level. Related references: PMID:11500381 PMID:11575918 PMID:18391071
- Functions as an E3 SUMO-protein ligase that catalyzes SUMO conjugation to substrate proteins, including viral immediate-early proteins, and exhibits robust isoform-dependent sumoylation activity (particularly isoforms PML-3 and PML-6). Related references: PMID:20972456 PMID:28250117
- Multiple isoforms (PML-1 to PML-14) display distinct subcellular localizations: nuclear isoforms (e.g., PML-4) localize to PML-NBs and nucleoplasm, while cytoplasmic isoforms (e.g., PML-14) associate with the endoplasmic reticulum and early endosome membranes, enabling context-dependent signaling in TGF-beta and other pathways.
- Key post-translational modifications include SUMOylation (essential for PML-NB formation), phosphorylation, and acetylation, which modulate its stability, interactions, and tumor-suppressive functions.
- Exhibits broad antiviral activity against both DNA and RNA viruses (e.g., herpesviruses, rabies, influenza, HCV) through isoform-specific mechanisms such as viral capsid sequestration, inhibition of viral mRNA/protein synthesis, and induction of apoptosis in infected cells.
- Plays a pivotal role in cellular senescence and apoptosis by positively regulating p53/TP53 stability and activity, and by repressing TBX2 transcription via the RBL2/E2F4 complex, linking PML to permanent growth arrest.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 300-600 of human PML (isoform 1), a central region expected to detect multiple isoforms; consider verifying reactivity in your specific model system due to alternative splicing.
- For Western blot, the predicted molecular weight is approximately 98 kDa, but isoforms and SUMOylated species may appear at higher molecular weights; using a high-quality protein ladder and optimizing transfer conditions is recommended.
- This antibody is validated for KO (knockout) confirmation; ensure to include appropriate KO control lysates in WB experiments to confirm band specificity.
- For IHC-P, antigen retrieval conditions (e.g., citrate buffer pH 6.0 or EDTA pH 9.0) should be optimized, and it is advisable to validate staining patterns against known PML-NB distribution in tissues.
- In ELISA applications, consider using the recombinant immunogen as a positive control to establish sensitivity and linear range.
