Biological Background: Alpha-synuclein Function and Localization
- Alpha-synuclein (SNCA), also known as NACP or PARK1, is a neuronal protein highly expressed in presynaptic terminals in the central nervous system.
- It plays a critical role in synaptic vesicle trafficking and neurotransmitter release by enhancing vesicle priming, fusion, and dilation of exocytotic fusion pores. Related references: PMID:28288128, PMID:30404828
- Mechanistically, it increases local Ca2+ release from microdomains to enhance ATP-induced exocytosis. Related references: PMID:30404828
- In its multimeric membrane-bound state, alpha-synuclein acts as a molecular chaperone that assists in the folding of SNARE proteins, crucial for sustaining SNARE-complex assembly during aging. Related references: PMID:20798282
- It also regulates dopamine neurotransmission by interacting with the dopamine transporter (DAT1) and modulating its activity. Related references: PMID:26442590
- Subcellularly, alpha-synuclein localizes to the cytoplasm, membrane, nucleus, synapse, and is secreted; it is also found in cell projections like axons.
- Mutations in SNCA are linked to Parkinson disease and other synucleinopathies, with alpha-synuclein accumulating in Lewy bodies.
Experimental Guidance and Technical Tips
- For Western blot, titrate the antibody to determine optimal dilution for your sample type; the expected band is approximately 14 kDa.
- For immunofluorescence, optimize fixation and permeabilization conditions suitable for synaptic protein detection.
- For ELISA, consider pairing this antibody with a detection antibody to develop a quantitative assay.
- The KO validation underscores specificity; use appropriate knockout controls to confirm target engagement.
- With confirmed cross-reactivity in mouse and rat, this antibody is suitable for studies in rodent models of neurodegeneration.
