Biological Background: MTH1 Function and Localization

  • MTH1 (NUDT1) is an oxidized purine nucleoside triphosphate hydrolase, also known as 8-oxo-dGTPase, that sanitizes the nucleotide pool to prevent DNA damage. Related references: PMID:10608900, PMID:12857738, PMID:22556419
  • It catalyzes the hydrolysis of 2-oxo-dATP into 2-oxo-dAMP and exhibits strong hydrolase activity toward 2-oxo-ATP, 8-oxo-dGTP, and 8-oxo-dATP. Related references: PMID:10373420, PMID:11139615
  • By removing oxidized purine nucleoside triphosphates, MTH1 prevents their incorporation into DNA and suppresses A:T to C:G and G:C to T:A transversion mutations. Related references: PMID:10373420, PMID:10608900, PMID:11756418
  • MTH1 also hydrolyzes methylated purine nucleoside triphosphates (e.g., O6-methyl-dGTP), blocking their integration into nucleic acids. Related references: PMID:30304478, PMID:32144205
  • Through these antimutagenic activities, MTH1 protects cells from oxidative stress-induced DNA damage. Related references: PMID:10608900, PMID:12857738, PMID:24695224
  • MTH1 localizes to the cytoplasm, cytosol, mitochondrial matrix, and nucleus, reflecting its role in sanitizing nucleotides in multiple compartments.
  • It is widely expressed, with highest levels in thymus, testis, embryo, and proliferating blood lymphocytes.
  • MTH1 is a magnesium-dependent hydrolase belonging to the Nudix family; it contains an alternative initiation site, a transit peptide for mitochondrial targeting, and has reported RNA-binding activity.

Experimental Guidance and Technical Tips

  • For Western blot, consider using whole cell lysates from human cell lines expressing endogenous MTH1; validate detection of the ~18 kDa band.
  • For immunoprecipitation, titrate the antibody amount and use appropriate negative controls.
  • Knockout validation confirms specificity; include a KO cell line as a negative control to ensure band identity.
  • For ELISA applications, test the antibody as a capture or detection reagent in a matched pair format.
