Biological Background: MLH1 Function and Localization

  • DNA mismatch repair protein Mlh1 (MutL protein homolog 1) is the catalytic subunit of the post-replicative DNA mismatch repair (MMR) system, forming the MutL alpha heterodimer with PMS2 and the MutL gamma heterodimer with MLH3.
  • MutL alpha is recruited to mismatched DNA by the MutS alpha (MSH2-MSH6) or MutS beta (MSH2-MSH3) complexes; it then activates PMS2 endonuclease to introduce strand breaks, initiating mismatch removal and repair.
  • MLH1 localizes predominantly to the nucleus and is associated with chromosomes during DNA replication and repair processes.
  • Expressed in multiple tissues including colon, lymphocytes, breast, lung, spleen, testis, prostate, thyroid, gall bladder, and heart.
  • Post-translational modifications include acetylation, phosphorylation, and ubiquitin conjugation, which may regulate its stability and function.
  • MLH1 is classified as a tumor suppressor; germline mutations are linked to hereditary nonpolyposis colorectal cancer (HNPCC), also known as Lynch syndrome.
