Biological Background: MAPKAPK3 Function and Localization
- MAP kinase-activated protein kinase 3 (MAPKAPK3), also known as chromosome 3p kinase (3pK), is a stress-activated serine/threonine-protein kinase encoded by the MAPKAPK3 gene in humans.
- It plays a key role in cytokines production, endocytosis, cell migration, chromatin remodeling and transcriptional regulation. Upon stress, it is phosphorylated and activated by p38-alpha/MAPK14, leading to substrate phosphorylation.
- MAPKAPK3 shares functional redundancy with MAPKAPK2, exhibiting the same substrate specificity, but its kinase activity and expression levels are lower. Both phosphorylate substrates such as HSP27/HSPB1, KRT18, KRT20, RCSD1, RPS6KA3, TAB3 and TTP/ZFP36.
- The protein localizes to both the nucleus and cytoplasm, enabling its involvement in diverse signaling processes.
- Widely expressed in tissues, with higher levels in heart and skeletal muscle; absent in brain. It is also expressed in the retinal pigment epithelium. Related references: PMID:26744326
- Involved in inflammatory responses by regulating TNF and IL6 production post-transcriptionally through phosphorylation of AU-rich element-binding proteins like TTP/ZFP36, modulating mRNA stability and translation.
- Required for acute TLR-induced macropinocytosis in dendritic cells via phosphorylation and activation of RPS6KA3, and also acts as a modulator of Polycomb-mediated repression.
- Post-translational modifications include phosphorylation and acetylation; the protein contains an ATP-binding kinase domain and is classified as a transferase.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-240) of human MAPKAPK3, which may contribute to recognition of the full-length protein (~43 kDa) in Western blotting. Consider including appropriate positive and negative controls in your experiment.
- Predicted molecular weight is 43 kDa; in WB, a band near this size is expected. Validate in your specific sample system as post-translational modifications or isoforms may cause slight shifts.
- For IF/ICC, note that MAPKAPK3 localizes to both nucleus and cytoplasm; optimize fixation and permeabilization conditions to preserve antigenicity. A nuclear counterstain is recommended.
- In ELISA, the recombinant immunogen (aa 1-240) can serve as a positive control; however, the antibody’s suitability for sandwich ELISA should be validated with paired antibodies.
- Cross-reactivity is predicted for human, mouse, and rat based on sequence homology; verify performance if used in other species.
