Biological Background: macroH2A.1 Function and Localization
- Core histone macro-H2A.1 is a variant histone H2A that replaces conventional H2A in a subset of nucleosomes, thereby repressing transcription. It compacts DNA into chromatin, limiting DNA accessibility for transcription, DNA repair, DNA replication, and chromosomal stability. - Inhibits binding of transcription factors (e.g., NF-kappa-B) and interferes with SWI/SNF remodeling complexes. Also inhibits histone acetylation by EP300 and recruits class I HDACs, inducing a hypoacetylated chromatin state. Related references: PMID:12718888, PMID:16428466, PMID:16107708
- Isoform 1 specifically binds poly-ADP-ribose and O-acetyl-ADP-ribose, playing a key role in NAD+ metabolism by capping PARP1-generated poly-ADP-ribose chains, thus limiting nuclear NAD+ consumption and promoting mitochondrial respiration.
