Biological Background: Lsh/HELLS Function and Localization
- Lsh/HELLS (Lymphoid-specific helicase), also known as SMARCA6, PASG, and ICF4, is an ATP-dependent chromatin remodeler of the SWI/SNF2 family.
- It plays a key role in epigenetic silencing by facilitating de novo DNA methylation at repetitive sequences, recruiting DNMTs and HDACs to form heterochromatin and silence transposable elements, thereby maintaining genomic stability.
Related references:
PMID:30307408 - In DNA damage response, HELLS is recruited to double-strand breaks in heterochromatin, mediating homologous recombination repair through RBBP8/CtIP-dependent end resection.
Related references:
PMID:22946062, PMID:31802118 - During meiosis, HELLS localizes to recombination hotspots by interacting with PRDM9, facilitating chromatin opening essential for meiotic recombination.
Related references:
PMID:32001511 - Subcellularly, HELLS localizes to the nucleus and associates with chromosomes, consistent with its chromatin remodeling functions.
- It is highly expressed in proliferative tissues such as thymus and testis, with lower expression in uterus, small intestine, colon, and peripheral blood mononuclear cells; also expressed in various neoplastic cell lines.
- Identified as a phosphoprotein, HELLS regulation likely involves phosphorylation; it also contains ATP-binding and coiled-coil domains.
- Mutations in HELLS are associated with immunodeficiency-centromeric instability-facial anomalies syndrome 4 (ICF4), highlighting its critical role in DNA methylation-dependent chromatin structure.
Experimental Guidance and Technical Tips
- This antibody was raised against a recombinant fusion protein spanning amino acids 719–838 of human HELLS (C-terminal region). For Western blotting, a band of approximately 97 kDa is expected; consider the possibility of post-translational modifications or isoforms that may alter migration.
- KO validation indicates that the antibody specifically recognizes HELLS; when using knockout cell lines, confirm loss of signal in the relevant experimental system.
- Applications include WB, IP, and ELISA. For IP, validate the enrichment of target protein in your sample system; for ELISA, appropriate coating and detection conditions should be optimized.
- Cross-reactivity is reported for human, mouse, and rat, making it suitable for studies across these species; however, verify reactivity in your specific tissue or cell type.
