Biological Background: LAMP2 Function and Localization
- LAMP2 (Lysosome-associated membrane glycoprotein 2) is a highly glycosylated type I transmembrane protein, also known as CD107b or LGP-96. Three main isoforms (LAMP-2A, -2B, and -2C) are produced by alternative splicing, each with distinct tissue distributions and roles in lysosomal function.
- It plays an essential role in lysosome biogenesis, lysosomal pH regulation, and autophagy. LAMP2 directly inhibits the proton channel TMEM175, facilitating lysosomal acidification required for optimal hydrolase activity.
