Lamins are intermediate filament proteins that form the nuclear lamina, a fibrous meshwork underlying the inner nuclear membrane, providing structural support and organizing chromatin. Lamin A and lamin C, both encoded by the LMNA gene via alternative splicing, are major components of this structure and are present in approximately equal amounts. In arteries, prelamin-A/C accumulates with age and disease, particularly in atherosclerotic lesions, colocalizing with senescent VSMCs. Expression increases due to oxidative stress–mediated downregulation of ZMPSTE24.
Experimental Guidance and Technical Tips
- The immunogen (aa 403–572 of human lamin A/C) targets a C-terminal region conserved between lamin A and lamin C, making this polyclonal antibody suitable for detecting both isoforms in human, mouse, and rat samples. The KO validation verifies specificity.
- For Western blotting, the expected molecular weight is ~74 kDa for lamin A; lamin C may appear around ~65 kDa. Consider loading adequate protein and using appropriate lysis conditions, as lamin solubility can be affected by ionic strength and detergent.
- In immunofluorescence and IHC, lamin A/C primarily stains the nuclear periphery (lamina). Permeabilization and fixation conditions should be optimized for the target cell type or tissue.
- For IP, the antibody may capture lamin A/C complexes; perform appropriate controls.
- As lamin A/C undergoes various post-translational modifications (e.g., phosphorylation, acetylation), treatment with inhibitors or induction of cell cycle arrest may alter detection.
