Biological Background: KRAS Function and Localization
- KRAS, also known as GTPase KRas, K-Ras 2, Ki-Ras, c-K-ras, or c-Ki-ras, is a member of the Ras family of small GTPases.
- It functions as a signal transducer in the Ras-MAPK signaling pathway, regulating cell proliferation and survival. Related references: PMID:22711838 PMID:23698361
- Ras proteins bind GDP/GTP and possess intrinsic GTPase activity. Related references: PMID:20949621 PMID:39809765
- Activates the MAPK1/MAPK3 cascade, leading to phosphorylation and ultimately degradation of GJA1.
- Promotes oncogenic events by inducing transcriptional silencing of tumor suppressor genes in colorectal cancer cells in a ZNF304-dependent manner. Related references: PMID:24623306
- Recognized by LZTR1, which mediates its ubiquitination by a BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complex. Related references: PMID:40934300
- Subcellularly localized to the cell membrane, endomembrane system, and cytoplasm.
- Expressed throughout the epidermis, but weakly in the stratum corneum and dermis at the protein level.
- Post-translational modifications include prenylation, palmitoylation, acetylation, methylation, and ubiquitination, contributing to its membrane association and regulation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-188 of human KRAS, covering the N-terminal region; consider this when interpreting detection of specific KRAS isoforms or mutants.
- For Western blot, a band near 22 kDa is expected; include positive and negative (knockout) controls to confirm specificity.
- In immunohistochemistry (IHC-P), validate staining in relevant tissue types with known KRAS expression, such as colorectal or pancreatic tissues.
- For ELISA, the antibody can be used as a capture or detection reagent; optimize concentrations for the specific assay format.
- Cross-reactivity with mouse and rat KRAS has been reported; verify performance in the intended species.
