Biological Background: KMT5C Function and Localization
- KMT5C (also known as SUV420H2) is a histone-lysine N-methyltransferase that specifically methylates monomethylated 'Lys-20' (H4K20me1) and dimethylated 'Lys-20' (H4K20me2) of histone H4 to produce di- and trimethylated forms, respectively; it also shows in vitro activity on unmethylated H4K20me0. - H4K20 trimethylation is a hallmark of transcriptional repression and constitutive heterochromatin formation, particularly at pericentric regions. KMT5C is targeted to histone H3 via interaction with RB1 family proteins (RB1, RBL1, RBL2).
- KMT5C facilitates TP53BP1 foci formation upon DNA damage and proficient non-homologous end-joining (NHEJ)-directed DNA repair by catalyzing the di- and trimethylation of H4K20. Related references: PMID:28114273

Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic peptide within amino acids 100–200 of human KMT5C. This linear epitope design makes it well-suited for denaturing western blotting (WB) and peptide-based ELISA.
- For WB, a band at approximately 52 kDa is expected. Use appropriate gel percentage and validate specificity by comparing signals in wild-type versus KMT5C knockout cell lysates.
- Cross-reactivity is confirmed for human and mouse; perform species-specific validation for other organisms.
- As a polyclonal antibody, note that inter-batch consistency may vary; consider lot-specific validation in critical assays.
