Biological Background: Ki67 Function and Localization
- Ki-67, encoded by the MKI67 gene, is also known as KIA, MIB-1, and PPP1R105. It is a large (~359 kDa) proline-rich nuclear protein that serves as a universal proliferation marker.
- Required to maintain individual mitotic chromosomes dispersed in the cytoplasm following nuclear envelope disassembly; acts as both a chromosome repellent during early mitosis and a chromosome attractant during late mitosis. Related references: PMID:27362226 PMID:32879492 PMID:35513709
- During early mitosis, relocalizes to the chromosome surface and forms extended brush-like structures that create a steric and electrostatic charge barrier, functioning as a surfactant to prevent chromosome aggregation and enable independent motility. Related references: PMID:27362226
- During anaphase, the brushes collapse and Ki-67 switches to a chromosome attractant: dephosphorylation exposes a basic patch, driving RNA-dependent liquid-liquid phase separation to cluster chromosomes and exclude large particles from the nascent nucleus. Related references: PMID:32879492 PMID:39153474 PMID:28935370
- Binds DNA with a preference for supercoiled and AT-rich sequences, potentially contributing to chromatin organization. Related references: PMID:10878551 PMID:24867636
- Localizes to chromosomes, nucleus, and nucleolus during the cell cycle.
- The protein undergoes acetylation, phosphorylation, and ubiquitination, which regulate its dynamic localization and function.
Experimental Guidance and Technical Tips
- For western blotting, Ki-67 is a high molecular weight protein (~359 kDa); use a low-percentage gel (e.g., 6% SDS-PAGE) and extended transfer times to ensure efficient detection.
- Validate the antibody in your specific experimental system, as Ki-67 expression is strictly cell-cycle dependent and may vary among cell types and proliferation states.
- For IHC-paraffin sections, standard antigen retrieval methods (heat-induced, pH 6 or 9) are recommended; expect nuclear or nucleolar staining in proliferating cells.
- The immunogen (aa 1200-1300) maps to the C-terminal region of human Ki-67; consider sequence alignment if cross-reactivity with other species is intended, as the antibody may recognize conserved epitopes.
