Biological Background: KEAP1 Function and Localization

  • KEAP1 (Kelch-like ECH-associated protein 1), also known as INrf2 or KLHL19, is a substrate-specific adapter for the BCR (BTB-CUL3-RBX1) E3 ubiquitin ligase complex.
  • It regulates oxidative stress response by targeting the transcription factor NFE2L2/NRF2 for ubiquitination and proteasomal degradation under normal conditions. Related references: PMID:14585973 PMID:15379550 PMID:15572695
  • Oxidative or electrophilic stress triggers covalent modification of reactive cysteine residues in KEAP1, inactivating the ligase and allowing NRF2 to accumulate and activate cytoprotective genes. Related references: PMID:16006525 PMID:17127771 PMID:18251510
  • KEAP1 also mediates ubiquitination of substrates like SQSTM1/p62, BPTF, and PGAM5, and is involved in selective autophagy pathways. Related references: PMID:15379550 PMID:17046835 PMID:20452972 PMID:28380357
  • Localizes to both cytoplasm and nucleus.
  • Broadly expressed, with highest levels in skeletal muscle.
  • Contains Kelch repeats and a BTB domain; classified under host-virus interaction, S-nitrosylation, and Ubl conjugation pathways.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 525-624) of human KEAP1, which includes Kelch repeat domains critical for substrate binding. This region is well conserved, supporting cross-reactivity with mouse and rat.
  • For Western blotting, a band around 70 kDa is expected. Consider that post-translational modifications (e.g., ubiquitination) or oxidative modifications may alter migration slightly.
  • For IF/ICC, both cytoplasmic and nuclear staining may be observed depending on cellular stress conditions.
  • The antibody has been KO validated, confirming specificity; however, always validate in your specific sample system and include appropriate controls.
  • For ELISA, direct binding to the immunogen peptide should be confirmed; pairing with a capture antibody may require optimization.
