  • ITCH is an E3 ubiquitin-protein ligase that accepts ubiquitin from E2 enzymes and directly transfers it to substrates, catalyzing Lys-29, Lys-48, and Lys-63-linked polyubiquitin chains to regulate protein stability, trafficking, and signaling.   • For Western blotting, a band near the predicted molecular weight of 103 kDa is expected. Use appropriate controls (e.g., ITCH knockout lysates or siRNA) to confirm specificity, especially given ITCH’s role as a large, multi-domain protein.
  • Cross-reactivity is indicated for human, mouse, and rat samples. Validate the antibody in your specific species and tissue context, as expression levels and post-translational modifications may affect recognition.
