Biological Background: ITCH Function and Localization
- ITCH is an E3 ubiquitin-protein ligase that accepts ubiquitin from E2 enzymes and directly transfers it to substrates, catalyzing Lys-29, Lys-48, and Lys-63-linked polyubiquitin chains to regulate protein stability, trafficking, and signaling. - As a core component of a ubiquitin-editing complex with TNFAIP3, TAX1BP1, and RNF11, ITCH promotes Lys-63 to Lys-48 ubiquitin switching on RIPK1, leading to its proteasomal degradation and termination of TNF/NF-κB inflammatory signaling. Related references: PMID:19131965

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1–155 of human ITCH (NP_001244066.1). This N-terminal region may allow detection of full-length ITCH and potential N-terminal-containing splice variants, but experimental verification in the relevant sample system is recommended.
- For Western blotting, a band near the predicted molecular weight of 103 kDa is expected. Use appropriate controls (e.g., ITCH knockout lysates or siRNA) to confirm specificity, especially given ITCH’s role as a large, multi-domain protein.
- Cross-reactivity is indicated for human, mouse, and rat samples. Validate the antibody in your specific species and tissue context, as expression levels and post-translational modifications may affect recognition.
- The antibody is suitable for ELISA; optimize capture/detection conditions using recombinant ITCH protein or cell lysates, and pair with a compatible detection system.
