Biological Background: IPO4 Function and Localization
- IPO4 (Importin-4), also known as Imp4b or RanBP4, is a nuclear transport receptor belonging to the importin-β family.
- It functions as a receptor for nuclear localization signals (NLS), mediating the nuclear import of diverse cargo proteins including histones, RPS3A, TNP2, and VDR in a Ran-dependent manner. Related references: PMID:11823430, PMID:16207705, PMID:17682055
- IPO4 specifically imports histone H3-H4 dimers when they are in complex with ASF1 chaperones, a critical step for chromatin assembly. Related references: PMID:21454524, PMID:29408485, PMID:36103578
- It also transports monomeric histones H3.1 and H4 into the nucleus. Related references: PMID:30177573
- IPO4 mediates the ligand-independent nuclear import of the vitamin D receptor (VDR), influencing transcriptional regulation. Related references: PMID:16207705
- In vitro, IPO4 can recognize a non-classical NLS on human cytomegalovirus UL84 protein to facilitate its nuclear entry. Related references: PMID:12610148
- Subcellularly, IPO4 shuttles between the cytoplasm and nucleus, consistent with its transport role.
- The protein is subject to acetylation and alternative splicing, and it contains HEAT repeats characteristic of importin-β family members.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 728–1081) of human IPO4; consider verifying that the antibody recognizes epitopes in this domain by testing in relevant cell or tissue lysates.
- For Western blotting, IPO4 is predicted at ~119 kDa; use an appropriate gel percentage and transfer conditions to ensure clear detection.
- The antibody shows cross-reactivity with mouse and rat IPO4; validate specificity and signal in your model system using positive and negative controls.
- KO validation suggests the antibody can be used for knockout/knockdown confirmation; include appropriate lysates from wild-type and IPO4-deficient samples when interpreting results.
- For ELISA applications, optimization of coating antigen and detection conditions is recommended to achieve optimal signal-to-noise ratios.
