Biological Background: IMPDH2 Function and Localization
- Catalyzes the conversion of inosine 5'-phosphate (IMP) to xanthosine 5'-phosphate (XMP), the first committed and rate-limiting step in de novo guanine nucleotide synthesis, thereby regulating cell growth. Related references: PMID:7763314 PMID:7903306
- Exhibits single-stranded nucleic acid-binding activity and may participate in RNA and/or DNA metabolism. Related references: PMID:14766016
- Implicated in the development of malignancy and the growth progression of certain tumors; expression profiling shows IMPDH2 predominates over IMPDH1 in tumor tissues.
- Localizes predominantly to the cytoplasm but can also be found in the nucleus and cytosol.
- Contains two CBS domains, functions as an oxidoreductase, and requires NAD and metal ions as cofactors.
- Underwent post-translational modifications including acetylation, phosphorylation, and ubiquitination, consistent with regulated protein turnover and signaling.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 100–300 of human IMPDH2, a region conserved across human, mouse, and rat, supporting the observed cross-reactivity. Sequence alignment may help predict reactivity with other species.
- For Western blotting, the expected band is approximately 56 kDa; consider using a KO-validated lysate pair as a stringent control to confirm band specificity.
- For immunofluorescence, typical nuclear and cytoplasmic staining should be expected; suitable fixation and permeabilization methods should be optimized.
- The antibody has been applied in ELISA; direct binding to the immunogen region suggests good suitability for quantitative assays, but dilution factors must be determined experimentally.
