Biological Background: hnRNP E1/PCBP1 Function and Localization

  • Poly(rC)-binding protein 1 (PCBP1), also known as hnRNP E1 or Alpha-CP1, is a member of the heterogeneous nuclear ribonucleoprotein (hnRNP) family.
  • PCBP1 functions as a single-stranded nucleic acid binding protein with high affinity for oligo dC sequences, regulating mRNA stability and translation. Related references: PMID:15731341 PMID:7556077 PMID:7607214
  • Together with PCBP2, PCBP1 is essential for erythropoiesis, likely through the regulation of mRNA splicing events during red blood cell development.
  • In the context of poliovirus infection, PCBP1 collaborates with the viral protein 3CD to initiate viral RNA replication.
  • PCBP1 localizes to both the nucleus and cytoplasm, reflecting its roles in RNA processing and transport.
  • Tissue expression is prominent in skeletal muscle, thymus, and peripheral blood leukocytes, with lower levels in prostate, spleen, testis, ovary, small intestine, heart, liver, adrenal, and thyroid glands.
  • Post-translational modifications of PCBP1 include acetylation, phosphorylation, isopeptide bond formation, and ubiquitin-like conjugation (Ubl), which may modulate its activity and interactions.
  • The protein has a molecular weight of approximately 37 kDa and contains conserved domains responsible for nucleic acid recognition and binding.

Experimental Guidance and Technical Tips

  • The antibody was raised against a peptide within the central region (aa 100-200) of human PCBP1; consider this when designing experiments that require N- or C-terminal epitope recognition.
  • For Western blot, endogenous PCBP1 is detected at ~37 kDa; include appropriate positive controls (e.g., HeLa, HEK293, or Jurkat lysates) and validate with knockout or knockdown if needed.
  • In IHC-P, optimize antigen retrieval conditions and validate staining specificity using known positive (e.g., skeletal muscle, thymus) and negative control tissues.
  • For ELISA, determine optimal coating concentration and antibody dilution using recombinant PCBP1 protein or peptide standard.
