Biological Background: Gasdermin-E Function and Localization

  • Gasdermin-E (GSDME/DFNA5/ICERE-1) is the precursor of a pore-forming protein that, upon cleavage by CASP3 or granzyme B, triggers pyroptosis, a form of programmed cell death distinct from apoptosis. Related references: PMID:27281216, PMID:28459430, PMID:32188940, PMID:33852854
  • The cleaved N-terminal fragment translocates to the plasma membrane, homooligomerizes, and forms pores of 10–15 nm diameter, facilitating the release of IL-1β and IL-18 and executing pyroptosis. Related references: PMID:28459430, PMID:32188940, PMID:33852854
  • GSDME mediates the switch from caspase-3-dependent apoptosis to pyroptosis in response to TNF, chemotherapy drugs, and viral infections, thereby promoting inflammatory cell death and anti-tumor immunity. Related references: PMID:27281216, PMID:28459430, PMID:32188940, PMID:28045099
  • As a tumor suppressor, GSDME is cleaved by granzyme B delivered by natural killer cells, inducing pyroptosis in target tumor cells and enhancing anti-tumor immune responses. Related references: PMID:21522185, PMID:31953257, PMID:32188940
  • GSDME also exhibits bactericidal activity and may play a role in p53/TP53-mediated DNA damage response. Related references: PMID:27281216, PMID:16897187
  • Expressed in multiple tissues, with highest expression in placenta and cochlea, consistent with its links to hearing loss and pregnancy-related pathologies. Related references: PMID:9771715
  • Post-translational modifications include palmitoylation and isopeptide bond formation; alternative splicing generates isoforms with varying molecular weights.
  • The full-length protein has a predicted molecular weight of ~55 kDa, while alternative splicing and cleavage produce fragments of ~11 kDa and ~35 kDa.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 245–344, which lies within the N-terminal region of GSDME. This region is critical for pore formation and is conserved in cleaved active fragments.
  • When performing Western blotting (WB), consider that multiple bands around 11 kDa, 35 kDa, and 55 kDa may be observed due to alternative splicing and proteolytic processing. Validate band identification with appropriate controls.
  • For immunohistochemistry (IHC-P), ensure antigen retrieval conditions are optimized for the detection of membrane-localized and cytosolic GSDME in formalin-fixed paraffin-embedded tissues.
  • In immunoprecipitation (IP) and ELISA applications, the monoclonal nature of this antibody provides high specificity, but cross-reactivity with other gasdermin family members should be ruled out using relevant knockout or siRNA controls.
