Biological Background: GIT1 Function and Localization

  • GIT1 (ARF GTPase-activating protein GIT1), also known as p95-APP1 and CAT1, is a multidomain scaffold protein that participates in receptor internalization, focal adhesion remodeling, and signaling by G protein-coupled receptors and tyrosine kinase receptors.
  • Through PAK1 activation, GIT1 positively regulates microtubule nucleation during interphase, and plays a role in cytokinesis via a pathway involving ENTR1 and PTPN13.

  • GIT1 promotes cell motility by regulating focal complex dynamics and locally activating RAC1. It also recruits MAPK1/3/ERK1/2 to focal adhesions after EGF stimulation, stimulating cell migration.

  • GIT1 is crucial for dendritic spine morphogenesis and synapse formation. In hippocampal neurons, it recruits ARHGEF7/beta-PIX to locally activate RAC1, essential for spine morphogenesis and synapse formation. It also regulates GABA(A) receptor synaptic stability by coordinating RAC1 and PAK1 activity, leading to F-actin stabilization and maintaining inhibitory synaptic transmission.
