Biological Background: ErbB3/HER3 Function and Localization
- ErbB3 (HER3) is a receptor tyrosine-protein kinase encoded by the ERBB3 gene, also known as proto-oncogene-like protein c-ErbB-3 and tyrosine kinase-type cell surface receptor HER3. Multiple isoforms exist due to alternative splicing (e.g., erbB3-S, p45-sErbB3, p85-sErbB3).
- Functions as a cell surface receptor for neuregulins, particularly neuregulin-1 (NRG1). Ligand binding induces tyrosine phosphorylation and promotes association with the p85 subunit of phosphatidylinositol 3-kinase (PI3K), initiating downstream signaling pathways. Experimental Guidance and Technical Tips
- The immunogen is a recombinant fragment corresponding to the C-terminus (aa 1275–1342) of human ErbB3. This region is common to full-length and certain secreted isoforms, so the antibody may detect multiple bands in WB.
- For WB, a band near 148 kDa is expected for the full-length receptor; consider reducing conditions and appropriate molecular weight markers.
- For IF/ICC, fixation and permeabilization may be required to access the intracellular epitope; validate localization in the appropriate cell line.
- KO validation indicates that the antibody exhibits reduced signal in ERBB3 knockout cells; consider using KO samples as negative controls to confirm specificity in your assay.
- Since it is a polyclonal antibody, lot-to-lot variability may exist; performance should be verified in your experimental system.
