Biological Background: EHMT2 Function and Localization
- EHMT2, also known as G9a, BAT8, or KMT1C, is a histone-lysine N-methyltransferase encoded by the EHMT2 gene (Gene ID: 10919) and belongs to the SET domain methyltransferase family.
- It functions as a euchromatic H3K9 mono- and dimethyltransferase (H3K9me1/H3K9me2), creating a repressive epigenetic mark read by HP1 proteins to silence transcription. Related references: PMID:11316813 PMID:20084102
- EHMT2 also monomethylates histone H3 at Lys-56 (H3K56me1) during G1 phase, facilitating PCNA interaction and DNA replication. Related references: PMID:22387026
- It also weakly methylates H3K27 and modifies histone H1.4 at Lys-26, expanding its histone substrate repertoire. Related references: PMID:11316813 PMID:19144645
- Beyond histones, EHMT2 methylates non-histone substrates including p53 (Lys-373), CDYL, WIZ, DNMT1, and HDAC1, and undergoes automethylation. Related references: PMID:20118233 PMID:18438403
- The protein is required for DNA methylation independently of its histone methyltransferase activity.
- EHMT2 localizes to the nucleus and chromosomes, consistent with its role in chromatin regulation.
- It is widely expressed with highest levels in fetal liver, thymus, lymph node, spleen, and peripheral blood leukocytes.
- Structurally, EHMT2 contains ankyrin repeats, a SET domain, and zinc finger motifs, classifying it as a metal-binding, S-adenosyl-L-methionine-dependent methyltransferase.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 877–1176 (C-terminal region) of human EHMT2; consider verifying reactivity in your specific model system as epitopes may be conserved across species or isoforms.
- For Western blot, the predicted molecular weight is approximately 132 kDa; use a high-range molecular weight marker and an appropriate gel percentage (e.g., 8% SDS-PAGE) for optimal resolution.
- In immunoprecipitation, test antibody concentration and incubation conditions (e.g., 1–2 µg per 500 µg lysate) to optimize capture efficiency and reduce background.
- For ELISA, coat the antigen at 1–10 µg/mL and titrate the antibody to determine the optimal working dilution; include appropriate negative controls.
- The supplier reports knockout validation for this antibody, supporting its specificity for endogenous EHMT2 detection; confirm target identity by siRNA knockdown or knockout cell lines when needed.
