Biological Background: EBP1/PA2G4 Function and Localization
- EBP1/PA2G4, encoded by the PA2G4 gene, is also known as proliferation-associated protein 2G4, ErbB3-binding protein 1, and cell cycle protein p38-2G4 homolog. It exists as multiple isoforms due to alternative splicing, with isoform 1 and isoform 2 being the predominant variants.
- It functions in ERBB3-regulated signal transduction and acts as a corepressor of the androgen receptor (AR). Its activity is regulated by the ERBB3 ligand neuregulin-1/heregulin. It inhibits transcription of some E2F1-regulated promoters, likely by recruiting histone acetylase activity.
- The protein binds RNA and associates with 28S, 18S, and 5.8S mature rRNAs, several rRNA precursors, and U3 small nucleolar RNA. It is involved in the regulation of intermediate and late steps of rRNA processing and may participate in ribosome assembly.
- EBP1/PA2G4 mediates cap-independent translation of specific viral IRESs and regulates cell proliferation, differentiation, and survival. Isoform 1 suppresses apoptosis, while isoform 2 promotes cell differentiation (By similarity).
- Subcellularly, it localizes to the cytoplasm, nucleus, and nucleolus.
- At the protein level, isoform 2 is undetectable in cancer cells, whereas isoform 1 is strongly expressed. Both isoforms are widely expressed in tissues including heart, brain, lung, pancreas, skeletal muscle, kidney, placenta, and liver.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitination. It is a phosphoprotein and undergoes isopeptide bond formation and Ubl conjugation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1–394 of human EBP1/PA2G4, which may detect both isoforms if the epitopes are conserved. Consider validating specificity using isoform-specific controls.
- For Western blot, expect a band near the calculated molecular weight of 44 kDa. Lysates from human, mouse, or rat cell lines can be used; cancer cell lines with high isoform 1 expression may provide strong signals.
- In IHC-P, optimization of antigen retrieval may be necessary. Validate staining patterns in known positive tissues, such as tumor sections.
- For ELISA applications, pair with an appropriate detection system. The KO validation data indicates that signal reduction in knockout cells can serve as a useful confirmatory assay.
