  • Mediates cellular responses to hypoxia, DNA damage, and energy depletion; its role in hypoxia-dependent mTORC1 inhibition is cell-type specific, occurring in fibroblasts and thymocytes but not hepatocytes.
  • Plays a role in p53/TP53-mediated apoptosis upon DNA damage by modulating mTORC1 activity, and is additionally required for antiviral defense by restricting viral protein synthesis.
  • Localizes to the mitochondrion and cytoplasm (primarily cytosol), consistent with its role in metabolic and stress signaling.
  • Broadly expressed across tissues with lower levels in brain, skeletal muscle, and intestine; protein levels are upregulated in substantia nigra neurons from Parkinson disease patients.
