Biological Background: Cytokeratin 8 Function and Localization

  • Keratin, type II cytoskeletal 8 (Cytokeratin-8, CK-8, KRT8) is a type II keratin that pairs with the obligate type I partner KRT18 to form intermediate filaments in simple epithelial cells.
  • Provides mechanical stability and resilience to epithelial cells and protects against non-mechanical stress such as apoptosis (By similarity).
  • Required for ARHGEF40-mediated actin stress fiber formation and tensional force-induced stress fiber formation and reinforcement (PubMed:26823019). Related references: PMID:26823019
  • In striated muscle, together with KRT19, may contribute to linking the contractile apparatus to dystrophin at the costameres (PubMed:16000376). Related references: PMID:16000376
  • Subcellular localization includes cytoplasm, nucleoplasm, nuclear matrix, and cytoskeleton.
  • Observed in epithelial tissues, muscle fibers accumulating in costameres, and in gingival mucosa and hard palate of the oral cavity.
  • Post-translational modifications include acetylation, glycosylation, isopeptide bond formation, methylation, phosphorylation, and ubiquitin-like conjugation.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 380-480 of human Cytokeratin 8. This region is likely conserved in mouse, explaining the observed cross-reactivity. Consider verifying epitope specificity when using in other species.
  • For Western blot applications, the predicted molecular weight is ~54 kDa, but additional bands around 56 kDa may appear due to post-translational modifications. Validate in relevant positive controls such as epithelial cell lysates.
  • This antibody has been validated for human and mouse samples. For other applications or species, appropriate validation is recommended.
