Biological Background: CSDE1 Function and Localization
- CSDE1, also known as Cold shock domain-containing protein E1, UNR (N-ras upstream gene protein), or D1S155E, is an RNA-binding protein containing cold shock domains. Alternative splicing generates multiple isoforms, contributing to its functional diversity.
- It is critically involved in translationally coupled mRNA turnover, particularly mediating the deadenylation and decay of FOS mRNA through the major coding-region determinant of instability (mCRD) domain in concert with other RNA-binding proteins. Related references: PMID:11051545 PMID:15314026
- CSDE1 is required for the efficient formation of cytoplasmic stress granules, which are sites of mRNA triage during cellular stress. Related references: PMID:29395067
- During microbial infection by human rhinovirus, CSDE1 is required for internal initiation of translation of viral RNA.
- The protein localizes to the cytoplasm and can be found in both stress granules and P-bodies, reflecting its dynamic role in mRNA metabolism.
- Post-translational modifications include acetylation, phosphorylation, and isopeptide bond formation (ubiquitination), which may regulate its activity and interactions.
- Structural studies have helped characterize the cold shock domain architecture of CSDE1, which is essential for RNA recognition and binding.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 369-600, located in the C-terminal region of human CSDE1. This region may encompass RNA-binding domains; thus, the antibody may recognize full-length and certain isoforms. Validate in your specific sample system.
- For Western blot (WB), a band of approximately 89 kDa is expected. KO validation confirms specificity; consider using a KO cell lysate as a negative control to verify band identity.
- In immunofluorescence (IF/ICC) and immunoprecipitation (IP), confirm subcellular localization and binding under relevant physiological conditions. Optimize fixation and permeabilization for IF.
- For ELISA, consider a standard indirect protocol and titrate both coating antigen and antibody concentration to determine optimal working conditions.
- Reported cross-reactivity includes human and mouse. For other species, verify sequence homology in the immunogen region before use.
