Biological Background: CSDE1 Function and Localization
- Cold shock domain-containing protein E1 (CSDE1), also known as UNR (N-ras upstream gene protein), is an RNA-binding protein with multiple cold shock domains.
- CSDE1 plays a critical role in translationally coupled mRNA turnover, particularly in the cytoplasmic deadenylation and decay of mRNAs such as FOS via the major coding-region determinant of instability (mCRD). Related references: PMID:11051545 PMID:15314026
- It is required for the efficient formation of stress granules, cytoplasmic foci that form under stress conditions. Related references: PMID:29395067
- CSDE1 localizes to the cytoplasm, and is enriched in stress granules and P-bodies, consistent with its role in mRNA regulation.
- The protein is subject to post-translational modifications including phosphorylation and acetylation, and is also known to undergo alternative splicing.
- During microbial infection, CSDE1 is required for internal initiation of translation of human rhinovirus RNA, highlighting its role in viral replication.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within the C-terminal half of CSDE1 (amino acids 369-600), which includes parts of the RNA-binding domains. Consider that this region may be conserved across species, supporting observed cross-reactivity with mouse.
- For Western blot, note the predicted molecular weight of approximately 89 kDa. Validate antibody specificity in the relevant sample system using appropriate controls.
- This polyclonal antibody is suitable for ELISA applications; consider optimizing coating conditions and detection reagents for your specific assay format.
