Biological Background: CHD1 Function and Localization
- CHD1 is an ATP-dependent chromatin-remodeling factor that acts as a substrate recognition component of the SAGA histone acetyltransferase complex, regulating RNA polymerase II transcription.
- It is also required for efficient transcription by RNA polymerase I, specifically the transcription termination step, and negatively regulates DNA replication.
- CHD1 helps maintain a specific chromatin configuration across the genome and is associated with histone deacetylase (HDAC) activity.
- The protein functions to bridge SNF2, FACT, PAF, and U2 snRNP complexes to trimethylated H3K4, modulating pre-mRNA splicing efficiency. Related references: PMID:18042460 PMID:28866611
- CHD1 is localized to both the nucleus and cytoplasm.
- It is widely expressed in many tissues, with highest levels in the brain, particularly the cerebellum and basal ganglia.
- CHD1 features chromodomains, a helicase ATP-binding domain, and a DNA-binding domain, and undergoes phosphorylation.
- Alternatively spliced isoforms exist.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide derived from the C-terminal region (aa 1600 to C-terminus); consider this when designing detection strategies.
- Validate antibody specificity in your experimental system, as polyclonal antibodies may exhibit lot-to-lot variability.
- For Western blot, the predicted molecular weight is approximately 197 kDa; use appropriate gel percentage and transfer conditions for high-molecular-weight proteins.
- Cross-reactivity with mouse and rat samples is reported; validate in the relevant sample system.
- For IHC-P, optimize antigen retrieval and antibody dilution.
- As CHD1 is involved in chromatin remodeling and splicing, this antibody may be suitable for studying transcription-related processes.
