Biological Background: CDKN1A/p21 Function and Localization
- CDKN1A/p21, also known as CIP1, WAF1, SDI1, MDA-6, and CAP20, is a key cyclin-dependent kinase inhibitor involved in cell cycle regulation.
- It plays a critical role in controlling cell cycle progression and DNA damage-induced G2 arrest, mediating p53/TP53-dependent and independent (via CREB3L1) inhibition of proliferation. Related references: PMID:9106657
- CDKN1A binds and inhibits cyclin-dependent kinase activity, preventing phosphorylation of critical substrates to block cell cycle progression. It also modulates cyclin D-CDK4 complex assembly and kinase activity in a dose-dependent manner.
- The protein inhibits DNA synthesis by competing with POLD3 for PCNA binding, disrupting DNA polymerase delta function. Related references: PMID:11595739
- In keratinocytes, it negatively regulates CDK4/6-driven phosphorylation of RB1, leading to release of E2F1 and transcription of G1/S phase genes.
- Localized in both the nucleus and cytoplasm, CDKN1A is ubiquitously expressed in adult tissues, with notably lower levels in the brain.
- Post-translational modifications include phosphorylation, acetylation, and ubiquitination, and it features a zinc-finger domain for metal binding.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (aa 1-164), which contains the CDK and PCNA binding domains, making this antibody a candidate for detecting functional interactions.
- For WB, consider that p21 migrates around 18-21 kDa; loading controls such as GAPDH or β-actin are recommended.
- Validate reactivity in mouse and rat samples, as cross-reactivity is reported; confirm band specificity in the species of interest.
- For IHC-P, optimize antigen retrieval and antibody dilution; both nuclear and cytoplasmic staining may be observed.
- ELISA applications may require titration of the antibody and antigen to achieve an optimal signal-to-noise ratio.
