Biological Background: CDK8 Function and Localization
- CDK8 (Cyclin-dependent kinase 8), also known as K35, IDDHBA, or Mediator complex subunit CDK8, is a serine/threonine-protein kinase and a core component of the Mediator complex.
- As a Mediator subunit, CDK8 functions as a transcriptional coactivator, bridging gene-specific regulatory proteins with the basal RNA polymerase II transcription machinery for regulated gene expression.
- CDK8 phosphorylates the C-terminal domain (CTD) of the large subunit of RNA polymerase II, which may inhibit the formation of a transcription initiation complex. Related references: PMID:10993082
