Biological Background: CDK2 Function and Localization
- Cyclin-dependent kinase 2 (CDK2), also known as p33 protein kinase or CDKN2, is a serine/threonine-protein kinase essential for cell cycle progression. It drives the G1-S transition and S phase progression by phosphorylating key substrates such as RB1, NPM1, and NPAT. Related references: PMID:10499802, PMID:10995386, PMID:11051553
- CDK2 is activated by cyclin E during early S phase and by cyclin A2 during late S phase/G2, regulating DNA replication, centrosome duplication, and histone gene transcription. Related references: PMID:10995387, PMID:11051553
- Its subcellular localization includes the nucleus, centrosome, Cajal body, cytoplasm, and endosome, reflecting its diverse roles in cell cycle control and DNA metabolism.
- CDK2 participates in DNA damage response and repair by phosphorylating BRCA2, NBN, and ERCC6, thereby modulating homologous recombination and telomere maintenance. Related references: PMID:15800615, PMID:20195506, PMID:21319273, PMID:28216226, PMID:29203878
- CDK2 phosphorylates the epigenetic regulator EZH2, promoting H3K27me3 maintenance and gene silencing, and also regulates the stability and activity of transcription factors like MYC and FOXP3. Related references: PMID:20935635, PMID:19966300
- Post-translational modifications of CDK2 include acetylation and S-nitrosylation, and its kinase activity requires ATP and magnesium. The calculated molecular weight of the human isoform is approximately 33.9 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 200–298) of human CDK2; consider this epitope location when designing blocking or competition experiments.
- For Western blotting (WB), a specific band around 34 kDa is expected; validate detection in a CDK2-expressing cell line (e.g., HeLa, HEK293) and confirm specificity using CDK2 knockout (KO) cell lysates if available.
- In immunohistochemistry-paraffin (IHC-P), antigen retrieval conditions (e.g., citrate buffer pH 6.0 or EDTA pH 9.0) may need optimization for your tissue type.
- ELISA applications may require titration of both capture and detection antibodies; consider using this polyclonal as part of a matched pair with a monoclonal anti-CDK2.
- Since this is a polyclonal antibody, verify lot-to-lot consistency for critical experiments, especially when transitioning between production batches.
