Biological Background: CD147/BSG (Basigin) Function and Localization
- Basigin (CD147/BSG) is a transmembrane glycoprotein and member of the immunoglobulin superfamily, also known as EMMPRIN, 5F7, TCSF, and HAb18G. It functions as a receptor and cell adhesion molecule.
- Acts as a retinal cell surface receptor for NXNL1, essential for normal retinal development and cone photoreceptor survival by enhancing aerobic glycolysis in photoreceptors (PubMed:25957687).

- Subcellular localization includes cell membrane (basolateral), melanosome, photoreceptor inner/outer segments, endosome, and endoplasmic reticulum membrane. Highly expressed in erythrocytes, melanoma cell lines, heart, kidney, skeletal muscle, testis, bone marrow, fetal liver, lung, and thymus; retina-specific expression in cone photoreceptors.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 45-200), enabling recognition of both native and denatured CD147. For WB, expect a band around 42 kDa, although glycosylation may cause slight shifts. KO validation confirms specificity; consider using appropriate positive and negative controls.
- For ELISA applications, the antibody may be used as either capture or detection reagent; evaluate optimal coating concentrations and pairing with matched detection antibodies in your assay system.
- Validate performance in biologically relevant sample types such as melanoma cell lines, erythrocyte lysates, or tissues with high CD147 expression (e.g., heart, kidney).
- Avoid repeated freeze-thaw cycles; store as recommended upon receipt (shipped on ice bag).
