Biological Background: Caspase-9 Function and Localization

  • Caspase-9 is a member of the cysteine-aspartic acid protease (caspase) family that functions as a key initiator caspase in the intrinsic apoptotic pathway. It is encoded by the CASP9 gene (GeneID: 842) and is also known as Apoptotic protease Mch-6, APAF-3, and ICE-LAP6.
  • Upon binding to Apaf-1, caspase-9 forms the apoptosome and undergoes activation. It then cleaves and activates downstream executioner caspases, primarily caspase-3 and caspase-7, to execute apoptosis.
  • Caspase-9 promotes DNA damage-induced apoptosis in an ABL1/c-Abl-dependent manner. It also proteolytically cleaves poly(ADP-ribose) polymerase (PARP) and BIRC6. Cleavage of BIRC6 occurs following inhibition of BIRC6-caspase binding by DIABLO/SMAC. Related references: PMID:36758105 PMID:36758106
  • An alternatively spliced isoform (isoform 2) lacks catalytic activity and acts as a dominant-negative inhibitor of caspase-9, providing a regulatory mechanism.
  • Tissue expression is ubiquitous, with highest levels in the heart (specifically in myocytes), moderate expression in liver, skeletal muscle, and pancreas, and low levels in other tissues.
  • As a zymogen, caspase-9 requires proteolytic cleavage for activation. It is further regulated by phosphorylation and ubiquitylation. The protein is classified as a thiol protease and its 3D structure has been resolved.

Experimental Guidance and Technical Tips

  • The immunogen used to raise this antibody covers amino acids 139–416, which includes the catalytic domain. Therefore, the antibody may detect both the full-length proform (~46 kDa) and cleaved active fragments. Consider including appropriate controls when assessing caspase activation.
  • For Western blotting, a primary band near 46 kDa is expected in untreated samples; additional smaller bands may appear upon induction of apoptosis. Validate the specific pattern in your model system.
  • This antibody has been KO validated, indicating reliable specificity for Caspase-9 in human, mouse, and rat samples. It is suitable for detecting endogenous levels.
  • In IF/ICC experiments, caspase-9 localizes predominantly to the cytoplasm but may be observed in the nucleus under certain apoptotic conditions. Optimize fixation and permeabilization protocols accordingly.
  • For ELISA applications, evaluate this antibody as part of a matched pair with other anti-Caspase-9 clones to achieve optimal sensitivity and specificity.
