Biological Background: Calnexin Function and Localization
- Calnexin (also known as IP90, p90, p88) is a calcium-binding, lectin-like chaperone of the endoplasmic reticulum (ER) that specifically interacts with newly synthesized monoglucosylated glycoproteins.
- It acts as a central quality-control component by retaining incorrectly folded or unassembled protein subunits in the ER, thereby facilitating proper protein folding and assembly.
- Calnexin is implicated in T-cell biology: it associates with incomplete T-cell antigen receptor complexes in immature thymocytes and is thought to contribute to thymocyte maturation signaling.
- Beyond the ER, Calnexin has been detected at the mitochondrion and melanosome membranes, and may participate in receptor-mediated endocytosis at synapses.
- The protein undergoes post-translational modifications including acetylation, phosphorylation, palmitoylation, disulfide bond formation, and ubiquitination, which may regulate its chaperone activity and localization.
Experimental Guidance and Technical Tips
- For Western blotting, expect a band near 68 kDa. Include appropriate positive and negative (KO) control lysates to verify specificity.
- The antibody is suitable for immunofluorescence/immunocytochemistry (IF/ICC) and immunohistochemistry (IHC-P) on human samples; optimize fixation and permeabilization protocols for your specific cell or tissue type.
- As the immunogen resides within the N-terminal region (aa 1-100), the antibody may detect N-terminal cleavage products or isoforms; consider running full-length controls.
- Since cross-reactivity is limited to human, carefully validate reactivity if applying to other species.
