Biological Background: Bax Function and Localization

  • BAX, also known as Bcl-2-like protein 4 (Bcl2-L-4), is a key regulator of the mitochondrial apoptotic pathway. Under stress conditions, it undergoes a conformation change, translocates to the mitochondrial outer membrane, and promotes the release of cytochrome c, which activates caspase-3 and triggers apoptosis. Related references: PMID:10772918 PMID:11060313 PMID:16113678
  • In normal cells, BAX is predominantly cytosolic due to continuous retrotranslocation from the mitochondria mediated by BCL2L1/Bcl-xL, which prevents its toxic accumulation at the mitochondrial outer membrane. Related references: PMID:21458670
  • Subcellularly, BAX localizes to the mitochondrial outer membrane, cytoplasm, and nucleus.
  • BAX is expressed in a wide variety of tissues. Isoform Alpha is found in spleen, breast, ovary, testis, colon, and brain, with lower levels in skin and lung. Isoform Sigma shows similar expression but is also detected in lung. Both isoforms are expressed in various cancer cell lines, including leukemia, lymphoma, breast, ovary, prostate, lung, and colon adenocarcinomas.
  • The BAX gene undergoes alternative splicing, giving rise to multiple isoforms, with Alpha and Sigma being the main functional variants.
  • Post-translational modifications include acetylation and ubiquitin conjugation. BAX is involved in tumor suppression and host-virus interactions.
  • The predicted molecular weight of BAX is approximately 21 kDa.

Experimental Guidance and Technical Tips

  • The immunogen for this antibody is a synthetic peptide corresponding to the N-terminal region (amino acids 1-100) of human Bax. This region is conserved across human, mouse, and rat, supporting the observed cross-reactivity.
  • For Western blotting, a specific band around 21 kDa is expected. Given its KO validation status, the antibody demonstrates high specificity and is suitable for detecting endogenous Bax levels.
  • When performing immunohistochemistry (IHC-P), standard antigen retrieval methods should be optimized, and appropriate controls are recommended to confirm staining specificity.
  • For ELISA applications, the antibody can be explored as either a capture or detection reagent; assay conditions should be optimized empirically.
  • Ensure proper validation in the relevant sample system, especially when using mouse or rat tissues, to account for any species-specific variations.
