Biological Background: ATG5 Function and Localization

  • ATG5, also known as autophagy protein 5, APG5-like, or apoptosis-specific protein, is a core component of the autophagy machinery. It is encoded by the ATG5 gene and is widely expressed in human tissues.
  • ATG5 is essential for autophagic vesicle formation. It conjugates with ATG12 via a ubiquitin-like system involving ATG7 (E1-like) and ATG10 (E2-like), forming the ATG12-ATG5 conjugate. This conjugate acts as an E3-like enzyme, facilitating the lipidation of ATG8 family proteins and their association with vesicle membranes.
  • The protein plays critical roles in mitochondrial quality control, cellular longevity, lymphocyte development, antigen presentation for MHC II, axon morphology maintenance, adipocyte differentiation, and primary ciliogenesis by removing OFD1 from centriolar satellites.
  • ATG5 is localized in the cytoplasm and at the preautophagosomal structure membrane, consistent with its function in autophagy.
  • ATG5 is involved in apoptosis, where its expression is a relatively late event downstream of caspase activity. It mediates IFN-gamma-induced autophagic cell death by interacting with FADD.
  • (Microbial infection) ATG5 can act as a proviral factor. It negatively regulates the innate antiviral immune response by impairing type I IFN production during vesicular stomatitis virus (VSV) infection (PubMed:17709747). Additionally, it is required for the translation of incoming hepatitis C virus (HCV) RNA and initiation of HCV replication (PubMed:19666601). Related references: PMID:17709747, PMID:19666601
  • The protein undergoes acetylation and is part of a ubiquitin-like conjugation system (isopeptide bond). It is associated with keywords including 3D-structure, alternative splicing, disease variant, host-virus interaction, and neurodegeneration.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1-275 of human ATG5 (NP_004840.1), which represents the full-length protein. This design likely captures the native conformation and may enhance recognition of endogenous ATG5.
  • The antibody is validated for WB, IF/ICC, IHC-P, and ELISA. For WB, a band at ~32 kDa is expected. Consider using ATG5 knockout controls to confirm specificity.
  • Given the high cross-reactivity with mouse and rat, this antibody can be used for studies in these model organisms.
  • For autophagy studies, induction of autophagic flux (e.g., starvation, mTOR inhibitors) may alter ATG5 conjugate levels; optimization of treatment conditions is recommended.
  • In apoptosis-related experiments, note that ATG5 expression increases downstream of caspase activity; time-course analysis may be necessary to capture changes.
  • For immunoprecipitation and IF/ICC, verify antibody compatibility with your buffer and fixation conditions, as ATG5 can be membrane-associated.
