Biological Background: ATG4A Function and Localization
- ATG4A is a cysteine protease that plays a key role in autophagy by mediating both proteolytic activation and delipidation of ATG8 family proteins. Related references: PMID:12473658, PMID:15169837, PMID:17347651
- It cleaves the C-terminal amino acid of ATG8 proteins (e.g., MAP1LC3A, GABARAP, GABARAPL2) to expose a glycine residue, enabling their conjugation to phosphatidylethanolamine (PE) and membrane insertion. Related references: PMID:12473658, PMID:15169837, PMID:17347651
- ATG4A also functions as a deubiquitinating-like enzyme that removes ATG8 from protein conjugates (ATG8ylation), counteracting high-molecular weight adducts. Related references: PMID:31315929, PMID:33773106
- It exhibits delipidation activity on PE-conjugated ATG8 proteins, a crucial recycling step during macroautophagy. Related references: PMID:29458288, PMID:33909989
- Compared to its homolog ATG4B, ATG4A shows stronger delipidation but weaker proteolytic processing activity. Related references: PMID:29458288
- Beyond its protease function, ATG4A promotes phagophore growth during mitophagy by regulating ATG9A trafficking and mediating ER–phagophore contacts. Related references: PMID:33773106
- ATG4A localizes primarily to the cytoplasm.
- Keywords: Alternative splicing, autophagy, hydrolase, protease, protein transport, lipid metabolism, Ubl conjugation pathway.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 289-398) of human ATG4A, which may help detect both full-length and potentially splice variants lacking N-terminal portions.
- For Western blot, the predicted molecular weight is 45kDa; ensure appropriate gel percentage and use positive control lysates from human or mouse cell lines.
- ELISA can be performed using recombinant ATG4A protein or peptide coating; consider checkerboard titration to optimize antibody and antigen concentrations.
- As a polyclonal antibody, lot-to-lot variability is possible; validate each new lot in the intended application.
- Knockout validation indicates specificity; include KO cell lysate as a negative control in WB to confirm band identity.
