Biological Background: ARHGEF1 Function and Localization
- ARHGEF1, also known as p115-RhoGEF, Sub1.5, or 115 kDa guanine nucleotide exchange factor, is a multidomain protein encoded by the ARHGEF1 gene.
- It acts as a bi-functional regulator of RhoA GTPase: functioning as a GTPase-activating protein (GAP) for GNA12 and GNA13, and as a guanine nucleotide exchange factor (GEF) for RhoA (PubMed:30521495, PubMed:8810315, PubMed:9641915).

Related references:
PMID:20098430 - In germinal center B cells, it participates in S1PR2-dependent signaling that inhibits growth and migration.
- Subcellularly, ARHGEF1 localizes to the cytoplasm and membrane.
- It is ubiquitously expressed across human tissues.
- The protein contains a coiled coil domain, undergoes alternative splicing, and is regulated by phosphorylation (keywords: Alternative splicing, Coiled coil, Phosphoprotein). 3D structures are available, and it has been identified in proteomics studies.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 783-912 of human ARHGEF1 (NP_004697.2). This C-terminal region may be conserved in known isoforms and rat orthologs, contributing to the observed cross-reactivity.
- For Western blot, the theoretical molecular weight is ~102 kDa. Use a validated positive control lysate from human or rat cell lines expressing ARHGEF1, and confirm band specificity with KO cell lysate if available.
- This antibody is suitable for IHC-P and ELISA. For IHC, optimize antigen retrieval conditions (e.g., heat-induced epitope retrieval in citrate buffer pH 6.0) and include appropriate tissue controls.
- As a polyclonal antibody, performance may vary between lots; consider validating each new lot for your specific assay requirements, especially for quantitative applications.
